I want a customer to make a one off payment using his debit card over the phone.
This will be a one off transaction as customers always pay by cheque or BACS.
What is the difference between opening a Virtual Terminal account and me just setting up a page with a buy now button for the amount outstanding and I enter my clients details that he will give me over the phone?

This would not be a good idea for security reasons, because when you enter credit card information from your own website PayPal's system can flag this transaction as fraud because you are entering your customers information on your own website. You can send an email invoice to your customer's email address, this will give them the option to pay with a credit card or PayPal account.
