Can I dispute the charge on my card if Paypal won't help?

So I purchased an item - I never received the item and opened a dispute through Paypal. The seller finally called me and said she would send it once I closed the dispute - which I did. STILL haven't received the item and I can't reopen the dispute with Paypal. Obviously I can dispute the charge with my bank, but does that effect my Paypal account in some way?

You should've never closed the dispute, the seller knew you couldn't reopen it. Yes use your funding source to reverse the charge, it won't affect your paypal acct, it will just be refunded.
