I just received a payment from Puerto Rico. Since this is my first sale how do I not know this is a scam?

What about this payment makes you think it's a scam? Sadly, there are scammers at work throughout the entire USA, but the legal resources to fight them are also present everywhere in the USA, including Puerto Rico. Dealing with a scammer in Puerto Rico is no different than dealing with one in Nevada, Delaware, Alaska or Louisiana. I lived in California and ran a business in Puerto Rico for several years without any problems.
