can I file a dispute on a item a bought from a website and if they don't respond I get a refund even if it was as described. Will they charge me?

If seller don't respond to the case in the dispute phase (first 20 days), escalate within the 20 days for PayPal to step in, if seller don't even respond to PayPal, PayPal may find in your favor. Responding in a timely manner is essential for either party or else.
Locate the transaction and click it and then click "report a problem". You should read the Purchase Protection terms to see how it works first, it may not cover every transaction/claim.
